We like staying in the centre of everything, so when we were in Rome we stayed right across an alley from the Fora and a couple blocks from the Colosseum. Hotel Nerva - super family service, decent breakfast, comfortable rooms. The Vatican tour is worth it - we booked with Angel Tours and had a solid tour guide. He made sure we got into the Sistine Chapel, while we watched other tour guides lead their groups outside, just because it was temporarily blocked until the chapel emptied out a little bit. In all of Italy, we were most impressed with Villa Borghese. Tremendous gallery with top Bernini sculptures - the plus is that entrance is by reservation and they only allow limited people in at a time - so you are not overrun and can spend time with the amazing art. And, of course, there are all the other sites to see, including the Colosseum, Trevi Fountain, Spanish Steps, Pantheon and so on.

Pre-book the Vatican museum for 8 am. You beat the crowds bc the regular line doesn't get in till 830. Run straight to the Sistine chapel and you'll get it to see it almost empty. That's what we did.
Agreed, Villa Borghese is a must and well worth it. Same thing, pick the first booking, and then don't linger in the foyer / lobby with all the mosaics, go straight to the 1st room and make your way through them. That way you'll get to see the beautiful Bernini's without a ton of ppl crowding around you. Come back to the foyer before you head to the 2nd floor.
Capotiline Museum is also phenomenal with stellar views of the forum. You get multiple re-entry I've a couple of days with that ticket so take advantage of it.
Book a tour for the coliseum from their own website. That way you skip the line entirely.
Go check out the small coliseum built by Hadrian near by, it's outstanding.
If you're into cocktails go to the Jerry Thomas Project. It's a speakeasy that was awesome! You have to head to their website to figure out the password for that day.

If you do the Collosseum tour in the morning you should be offered an afternoon tour of Palatine Hill. EXCELLENT. Also, really enjoyed Castel St Angelo.
